Cell culture is a complex process by which cells, prokaryotic or eukaryotic, are cultured under controlled conditions, usually outside their natural environment.

There are essentially 2 types of cell cultures :cell line generation and primary culture generation

  • The generation of primary cells, which consists in isolating cells directly from the tissues of living organisms. These cells are cultured in vitro and have a limited number of cell divisions.
  • The generation of the immortalized cell line, the cell lines have escaped cell senescence and can therefore divide indefinitely, by successive transplantation.

Functional cellular models allow compounds to be profiled by looking at their impact on defined targets.

Immortalized cells are used as research models in biochemistry and cell biology. Primary cultures, on the other hand, are typically used as models of physiological state in vivo. There are indeed different possible tests on cells.

Cell lines generation

You have the possibility to generate cell lines, these can be stable or immortalized lines, primary cultures or even pluripotent stem cell lines (IPS or HIPSC). IPS are stem cell lines generated from already differentiated adult cells, which can divide indefinitely. The creation of HIPSCs is done by transducing "stem" genes using a viral system.

In vitro cellular measurements and tests

Different tests are possible on the cells, our Experts allow you to look at In vitro cellular measurements and testscell migration and invasion through chemotaxia, haptotaxis, transmigration, using a Boyden chamber or a selection of 2D-gap closure tests. The study of cell migration can also be done in 3D environments, allowing the extracellular matrix to be simulated. You can also perform viability tests (MTT or XTT tests, the measurement of[ATP]), proliferation (measurement of DNA synthesis (thymidine-3H or BrdU), metabolic activity (XTT or MTT), adhesion, apoptosis (measurement of caspase activity, cytochrome C release, nucleus marking, TUNEL,...).

It is also possible to carry out oxidative stress tests (ROS measurement, catalase activity test, lipid peroxidation, etc.).

In addition, other more specific tests are possible such as cell transfection, the addition of compounds and hormones, the use of different culture conditions (temperature, 02, C02, medium) or the expression of protein under radiolabelled conditions.

The results of the tests can be obtained at several times and in different formats: cells, lysates, imaging.

Purification of cellular organelles 

Our experts offer services for the purification of mitochondria, endoplasmic reticulum, nucleus, proteasomes, peroxisomes and ribosomes.

Depending on the cellular component, purification can be done by centrifugation, magnetic beads, fractionation, successive centrifugations or affinity purification.

Depending on the organelle, different degrees of purification can be obtained (raw RE; fraction enriched in rough RE, fragment of enriched raw peroxisome,...).

You also have the possibility to perform a gradient density separation, the separation conditions can be fully defined, in a custom format, to ensure the enrichment of important fractions for a given project.

Cell sorting

Cell sorting makes it possible to select and isolate several cells according to selection criteria. Single cell sorting is the first step before using new techniques and technologies such as single cell sequencing, qPCR and digital PCR. Sorted cells can in some cases be used for future uses, such as re-culture.
